Summary

Activated bleomycin cleaves transfer RNA (tRNA) specifically, releasing adenine and uracil. This drug also binds to yeast tRNA, similar to its interaction with DNA.

Area of Science:

  • Biochemistry
  • Molecular Biology
  • Pharmacology

Background:

  • Bleomycin is a known DNA-cleaving agent.
  • The interaction of bleomycin with RNA has not been previously characterized.

Purpose of the Study:

  • To investigate the effect of activated bleomycin on transfer RNA (tRNA).
  • To determine the specificity and mechanism of bleomycin-induced tRNA cleavage.

Main Methods:

  • Incubation of yeast tRNAPhe with activated bleomycin and Fe(III)-bleomycin.
  • Analysis of reaction products to identify released nucleobases.

Main Results:

  • Activated bleomycin specifically cleaves yeast tRNAPhe in a dose-dependent manner.
  • Adenine and uracil are released as products of tRNA cleavage.
  • Bleomycin and Fe(III)-bleomycin bind to yeast tRNAPhe, similar to their binding with B-DNA.

Conclusions:

  • Bleomycin exhibits sequence-specific cleavage activity against tRNA.
  • The findings suggest a potential new mechanism of action for bleomycin and offer insights into drug-RNA interactions.
