1Koçarlı Vocational and Training School, Adnan Menderes University, Aydın, Turkey.
Summary
This study developed a novel cryogel functionalized with Cibacron Blue F3GA dye for immobilizing alcohol dehydrogenase. The dye-attached cryogel demonstrated efficient and reversible protein immobilization with high reusability.
Area of Science:
- Biomaterials Science
- Biotechnology
- Polymer Chemistry
Background:
- Enzyme immobilization is crucial for biocatalysis and diagnostics.
- Developing efficient and reusable immobilization matrices remains a challenge.
Purpose of the Study:
- To synthesize and characterize a poly(HEMA-co-GMA) cryogel functionalized with Cibacron Blue F3GA.
- To investigate the reversible immobilization of alcohol dehydrogenase (ADH) using the functionalized cryogel.
Main Methods:
- Cryopolymerization technique for poly(HEMA-co-GMA) cryogel synthesis.
- Functionalization with Cibacron Blue F3GA dye.
- Characterization using ESEM, EDX, and swelling studies.
- Investigation of ADH adsorption under varying pH, concentration, temperature, and ionic strength.
Main Results:
- The cryogel exhibited a porous surface morphology (30-50 μm pores).
- Maximum ADH adsorption capacity reached 8.55 mg/g cryogel.
- Successful desorption and reuse of the cryogel for 25 cycles with minimal capacity loss.
Conclusions:
- The Cibacron Blue F3GA-functionalized cryogel is an effective matrix for reversible alcohol dehydrogenase immobilization.
- The developed material shows high stability and reusability, suitable for biotechnological applications.
