This study investigated increasing Nimesulide solubility using Tween 80. Results showed complex solubility behavior, with low surfactant concentrations potentially decreasing Nimesulide solubility, requiring further investigation.
Area of Science:
- Pharmaceutical Sciences
- Drug Delivery
- Physical Chemistry
Background:
- Nimesulide, a non-steroidal anti-inflammatory drug (NSAID), exhibits poor aqueous solubility (≈ 0.01 mg/mL).
- As a BCS 2 drug (poorly soluble, highly permeable), Nimesulide presents formulation challenges impacting bioavailability.
- Surfactant-based solubilization above critical micelle concentration (CMC) is a strategy for poorly soluble drugs.
Purpose of the Study:
- To enhance the aqueous solubility of Nimesulide using varying concentrations of Tween 80 in a pH 7.4 buffer.
- To understand the relationship between Tween 80 concentration and Nimesulide solubility for improved pharmaceutical formulation.
Main Methods:
- Solubilization of Nimesulide in aqueous buffer (pH 7.4) with different concentrations of Tween 80.
- Analysis of Nimesulide solubility and release characteristics at various surfactant concentrations.
Main Results:
- Nimesulide solubility dependence on Tween 80 concentration was non-linear.
- A decrease in apparent solubility was observed at very low Tween 80 concentrations, possibly linked to CMC.
- Potential analytical artifacts, such as Nimesulide precipitation or colloidal formation, require further investigation.
Conclusions:
- Tween 80 can increase Nimesulide solubility, but the relationship is complex and concentration-dependent.
- The observed non-linear behavior and potential precipitation at low concentrations necessitate careful formulation design.
- Further research is needed to elucidate the mechanisms behind Nimesulide-Tween 80 interactions and optimize formulation strategies.
