    Area of Science:

    • Immunology
    • Pharmacology
    • Traditional Chinese Medicine

    Background:

    • Asthma is a chronic respiratory disease characterized by airway inflammation.
    • The balance of T helper 1 (Th1) and T helper 2 (Th2) cytokines plays a crucial role in asthma pathogenesis.
    • Stemona tuberosa is a traditional herb used for respiratory ailments, but its processed form's efficacy requires further investigation.

    Purpose of the Study:

    • To investigate the therapeutic effects of crude and processed Stemona tuberosa ethanol extracts on OVA-induced asthma in mice.
    • To compare the immunomodulatory effects of crude versus processed S. tuberosa on Th1/Th2 cytokine levels and T-bet/GATA-3 gene expression.
    • To determine the pharmacological differences between crude and processed S. tuberosa and their clinical implications.

    Main Methods:

    • Establishment of an OVA-induced asthma mouse model.
    • Oral administration of different concentrations of crude and processed Stemona tuberosa ethanol extracts.
    • Measurement of Th1 (INF-gamma) and Th2 (IL-5) cytokine levels in splenocytes.
    • RT-PCR analysis to determine T-bet/GATA-3 gene expression in lung tissue.

    Main Results:

    • Processed S. tuberosa significantly increased INF-gamma and decreased IL-5 levels in splenocytes compared to crude extracts (P < 0.05).
    • Administration of processed S. tuberosa notably increased the T-bet/GATA-3 ratio in lung tissue (P < 0.05).
    • Both crude and processed extracts demonstrated asthma treatment potential by regulating the Th1/Th2 ratio, with processed samples showing a more significant effect.

    Conclusions:

    • Ethanol extracts of both crude and processed Stemona tuberosa can treat asthma by modulating the Th1/Th2 immune balance.
    • Processed Stemona tuberosa exhibits superior therapeutic efficacy for asthma compared to crude extracts.
    • Processed S. tuberosa is more suitable for clinical application in treating asthma and chronic cough, warranting further research into its processing mechanisms.
