Natural Killer (NK) cell function is not altered in ankylosing spondylitis (AS) patients, regardless of HLA-B27 status. However, NK cell activity was significantly decreased in AS patients taking NSAIDs.

Area of Science:

  • Immunology
  • Rheumatology
  • Cell Biology

Background:

  • Ankylosing spondylitis (AS) is a chronic inflammatory disease primarily affecting the axial skeleton.
  • The role of Natural Killer (NK) cells, crucial components of the innate immune system, in AS pathogenesis remains unclear.
  • Previous research has not established a clear link between NK cell function and AS, nor its association with the HLA-B27 gene.

Purpose of the Study:

  • To investigate the function of Natural Killer (NK) cells in patients with ankylosing spondylitis (AS).
  • To determine if NK cell activity differs between AS patients and healthy controls.
  • To explore the potential influence of HLA-B27 status and non-steroidal anti-inflammatory drug (NSAID) treatment on NK cell function in AS.

Main Methods:

  • Evaluated NK cell function in 28 AS patients (21 HLA-B27 positive) and controls.

Related Experiment Videos

  • Utilized a Leu7 (HNK-1) monoclonal antibody for NK cell identification.
  • Assessed spontaneous NK cell cytotoxicity against K562 target cells using a flow cytometric assay.
  • Main Results:

    • No significant difference in NK cell activity was observed between AS patients and controls.
    • NK cell function did not differ between HLA-B27 positive and negative AS patients.
    • A significant decrease in NK cell activity was found in AS patients undergoing NSAID treatment compared to those not treated.

    Conclusions:

    • The study found no evidence that HLA-B27 status influences NK cell function in AS.
    • NSAID treatment, contrary to expectations, was associated with decreased NK cell activity in AS patients.
    • The observed reduction in NK cell activity may reflect disease progression or play a role in AS pathogenesis, potentially related to bacterial antigen persistence.
