The NQO1 C609T polymorphism shows no significant link to prostate cancer risk overall. However, this genetic variation may increase prostate cancer susceptibility in Asian populations, warranting further investigation.
Area of Science:
- Genetics
- Oncology
- Epidemiology
Background:
- Conflicting results exist regarding the association between the NQO1 C609T polymorphism and prostate cancer risk.
- The NAD(P)H:quinone oxidoreductase 1 (NQO1) enzyme plays a role in carcinogen detoxification.
Purpose of the Study:
- To systematically review and meta-analyze published studies to assess the association between the NQO1 C609T polymorphism and prostate cancer risk.
- To investigate potential ethnic differences in this association.
Main Methods:
- A meta-analysis was conducted on 6 eligible studies comprising 717 cases and 1,794 controls.
- Studies were identified from PubMed and the Cochrane Library.
- Statistical analyses were performed using Review Manager 5.0 and Stata 11.0.
Main Results:
- Overall, no significant association was found between the NQO1 C609T polymorphism and prostate cancer risk across additive, recessive, dominant, and allelic models.
- Stratified analysis by race indicated a significant association in Asians under dominant and allelic models (OR = 1.419, P = 0.021; OR = 1.337, P = 0.040, respectively).
- No significant associations were observed in Caucasians.
Conclusions:
- The NQO1 C609T polymorphism may contribute to increased prostate cancer susceptibility in Asian populations.
- Further large-scale, well-designed case-control studies are required to validate these findings.
