Fragile X premutation carriers show higher rates of macroorchidism and lower IQ scores. This study links macroorchidism to reduced verbal and full-scale IQ in these males.
Area of Science:
- Genetics
- Neuroscience
- Endocrinology
Background:
- Fragile X syndrome is characterized by macroorchidism and intellectual disability, linked to reduced Fragile X Messenger Ribonucleoprotein (FMRP) levels.
- FMRP is highly expressed in the brain and testes, but the relationship between macroorchidism and intellectual disability in premutation carriers is unstudied.
Purpose of the Study:
- To investigate the prevalence of macroorchidism in males with Fragile X premutation.
- To examine the association between macroorchidism and IQ in premutation carriers.
Main Methods:
- Evaluated macroorchidism status in 213 males (142 premutation carriers, 71 controls).
- Assessed the association between macroorchidism, FMR1 mRNA levels, CGG repeat number, age, and IQ (verbal and full scale).
Main Results:
- Macroorchidism prevalence was significantly higher in premutation carriers (32.4%) than controls (5.6%).
- Increased FMR1 mRNA levels correlated with a higher odds ratio for macroorchidism in carriers.
- Premutation carriers with macroorchidism exhibited lower verbal and full-scale IQ scores compared to those without.
Conclusions:
- Macroorchidism is more prevalent in Fragile X premutation carriers.
- A correlation exists between macroorchidism and lower verbal and full-scale IQ in premutation carrier males.
- Further research is needed to determine if elevated FMR1 mRNA or reduced FMRP levels underlie these associations.
