Synthesis and Regulation of Thyroid Hormones01:20

Synthesis and Regulation of Thyroid Hormones

7.1K
Low blood levels of the thyroid hormones — triiodothyronine (T3) and thyroxine (T4) — signal the hypothalamus to release the thyrotropin-releasing hormone (TRH). TRH then reaches the pituitary gland and stimulates the release of thyroid-stimulating hormone(TSH) into the bloodstream.
Upon reaching the thyroid gland, TSH stimulates the follicular cells' active uptake of iodide ions from the blood. The ions diffuse to the apical surface of the cells and are oxidized to iodine. The...

The Thyroid Gland01:23

The Thyroid Gland

6.9K
The thyroid gland is a small, butterfly-shaped gland located in the neck and covers the anterior surface of the trachea. The gland has two lateral lobes connected by a thin tissue mass called the isthmus. Internally, each lobe comprises many small spherical structures known as thyroid follicles, surrounded by a network of blood vessels.
The follicles have a central cavity lined by simple cuboidal to squamous epithelial cells called follicular cells. These cells produce the glycoprotein...

Summary

This study presents a novel mouse embryonic thyroid explant culture system that mimics in vivo development over four days. This method allows researchers to study thyroid organogenesis, including angio-follicular unit formation and cell differentiation.

Area of Science:

  • Developmental Biology
  • Endocrinology
  • Cell Biology

Background:

  • Thyroid hormones (T3, T4) and calcitonin are crucial for development and metabolism.
  • The intricate formation of angio-follicular units and cell interactions in the thyroid remains poorly understood.
  • Existing knowledge of adult thyroid structure and function contrasts with gaps in understanding early organogenesis.

Purpose of the Study:

  • To develop and validate a mouse embryonic thyroid explant culture system for studying organogenesis.
  • To investigate the formation of angio-follicular units and cell differentiation during thyroid development.
  • To provide a tool for manipulating and analyzing thyroid development ex vivo.

Main Methods:

  • Dissection and culture of mouse embryonic thyroid anlagen on semiporous filters or slides.
  • Observation of explant development over four days, recapitulating in vivo processes.
  • Analysis of explants using real-time imaging, immunostaining, and RT-qPCR.

Main Results:

  • The culture system successfully replicates key in vivo thyroid developmental events, including bilobation, follicle formation, cell differentiation, and vascularization.
  • Thyrocytes and C-cells differentiate, and endothelial cells proliferate, migrate, and associate with epithelial cells.
  • The system supports development from wild-type, knockout, and transgenic embryos, allowing for genetic manipulation.

Conclusions:

  • Thyroid explant culture is a powerful ex vivo system for studying thyroid organogenesis and differentiation.
  • This method enables manipulation and detailed analysis of morphogenetic events, angio-follicular unit formation, and cell communication.
  • The system facilitates research into the complex processes governing thyroid gland development.
