Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

2.9K
Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl...

Mechanism of Angiogenesis01:10

Mechanism of Angiogenesis

6.3K
Blood vessel formation starts early during embryonic development, around day 7. In the extraembryonic yolk sac, mesodermal precursor cells called hemangioblast proliferate and differentiate into angioblast. Angioblasts express vascular endothelial growth factor receptor 2 or VEGFR2, which binds VEGF-A, a proangiogenic factor, guiding blood vessel formation. VEGF signaling promotes angioblasts to form a blood island in the developing embryo. Angioblasts further differentiate, giving rise to...

Area of Science:

  • Marine biotechnology
  • Carbohydrate chemistry
  • Molecular pharmacology

Background:

  • Fucoidans are sulfated polysaccharides found in brown algae, known for diverse biological activities.
  • Their structural heterogeneity influences biological functions, particularly angiogenesis.

Purpose of the Study:

  • To review recent findings on the structural variability of fucoidans.
  • To summarize their anti- and pro-angiogenic properties.

Main Methods:

  • Literature review of recent publications on fucoidan structure and function.
  • Analysis of structure-activity relationships concerning angiogenesis.

Main Results:

  • Fucoidans show considerable structural differences in backbone, branches, monosaccharide composition, and sulfation.
  • The degree of sulfation and molecular weight critically influence fucoidan's biological effects on angiogenesis.
  • High-molecular-weight fucoidans display anti-angiogenic activity, while low-molecular-weight fractions promote angiogenesis.

Conclusions:

  • Fucoidan's biological activity, especially in angiogenesis, is strongly linked to its structural characteristics.
  • Molecular weight and sulfation are key determinants of fucoidan's pro- or anti-angiogenic effects.
  • Further research is needed to elucidate the impact of other structural details on fucoidan's role in angiogenesis.
