Drug Dosing in Renal Diseases: Estimation of Glomerular Filtration Rate Based on Serum Creatinine Concentration

337
Glomerular filtration rate (GFR) can be estimated from serum creatinine using the modification of diet in renal disease (MDRD) formula or the chronic kidney disease–epidemiology collaboration (CKD–EPI) equation. Both methods are widely used in clinical practice to assess kidney function and guide treatment decisions. The MDRD equation does not require weight or height measurements and is normalized to the body surface area of 1.73 m², considered the average adult surface area.

Area of Science:

  • Genomics
  • Bioinformatics
  • Renal Medicine

Background:

  • Limited therapeutic and preventive options exist for renal failure.
  • Gene expression profiling can identify biological differences between patients and healthy individuals.
  • Previous analyses often overlooked biological and environmental factors.

Purpose of the Study:

  • To investigate gene expression differences in end-stage renal failure patients compared to healthy controls.
  • To utilize partial least squares (PLS) analysis for identifying differentially expressed genes.
  • To explore molecular signatures associated with renal failure.

Main Methods:

  • Gene expression data was sourced from the Gene Expression Omnibus database.
  • Partial least squares (PLS) analysis was employed to identify differentially expressed genes.
  • Enrichment and network analyses were conducted to understand molecular pathways.

Main Results:

  • 573 differentially expressed genes were identified.
  • Enrichment analyses revealed dysregulation in various biological processes.
  • Network analysis highlighted seven hub genes, including CDK2, TP53, and RELA, linked to renal failure progression.

Conclusions:

  • The study elucidates the gene expression characteristics of renal failure patients.
  • Identified molecular signatures may serve as potential therapeutic targets.
  • Further research can leverage these findings for novel treatment strategies.
