Area of Science:

  • Pharmacology
  • Dermatology

Background:

  • Nitroglycerin is used to treat angina.
  • Transdermal drug delivery offers potential advantages over oral administration.

Purpose of the Study:

  • To compare the transdermal absorption of nitroglycerin from a novel adhesive patch versus a conventional ointment.
  • To evaluate the pharmacokinetic parameters and delivery efficiency of the new nitroglycerin patch.

Main Methods:

  • A two-period crossover study involving 24 healthy men.
  • Comparison of a 20-cm2 nitroglycerin patch (q24hr) with 2% nitroglycerin ointment (q8hr) over a 50-cm2 area.
  • Analysis of plasma pharmacokinetic parameters and residual drug content.

Main Results:

  • The nitroglycerin patch delivered approximately 1.5 times more drug per unit surface area compared to the ointment.

Related Experiment Videos

  • Nitroglycerin release rate from the patch was determined to be 0.75 mg/cm2/24 hrs.
  • The transdermal patch demonstrated excellent skin adhesion throughout the 24-hour application period.
  • Conclusions:

    • The new nitroglycerin adhesive transdermal patch provides efficient drug delivery.
    • The patch is a viable alternative for transdermal nitroglycerin administration with improved delivery compared to ointment.
    • Further studies may explore the clinical efficacy of this new patch formulation.
