T300A genetic polymorphism: a susceptibility factor for Crohn's disease?

This study investigated the T300A polymorphism in the ATG16L1 gene and its association with Crohn's disease risk. The results did not confirm a significant link between this genetic variation and increased susceptibility to Crohn's disease.

- Genotyping of the T300A polymorphism in the ATG16L1 gene was performed using Polymerase Chain Reaction and Restriction Fragment Length Polymorphism analysis.
- DNA samples from 106 Crohn's disease patients and 238 healthy controls from Southern Brazil were analyzed.
- Agarose gel electrophoresis was used to separate and visualize amplified DNA fragments.
Main Results:
- The AG genotype was the most prevalent in both Crohn's disease patients (50%) and controls (44.8%).
- Genotype frequencies for AA were 26.4% in patients vs. 35.1% in controls, and GG were 23.6% in patients vs. 20.1% in controls.
- The frequency of the G allele was higher in Crohn's disease patients (48.6%) compared to controls (42.4%), but this difference was not statistically significant.
Conclusions:
- The study did not find statistically significant evidence to confirm that the T300A polymorphism in the ATG16L1 gene confers increased susceptibility to Crohn's disease.
- Further research with larger cohorts may be needed to fully elucidate the role of this polymorphism in Crohn's disease pathogenesis.
