Area of Science:

  • Natural Product Chemistry
  • Pharmacology

Background:

  • Artemisia absinthium is a plant known for its traditional medicinal uses.
  • Dimeric guaianolides are a class of natural compounds with diverse biological activities.

Purpose of the Study:

  • To isolate and characterize dimeric guaianolides from Artemisia absinthium.
  • To evaluate the inhibitory potential of these compounds on nitric oxide production.

Main Methods:

  • Isolation of compounds using chromatographic techniques.
  • Structure elucidation via 1D/2D NMR and HRESIMS.
  • Determination of absolute configuration using X-ray crystallography.
  • Biological activity assessment using BV-2 cell-based assays.

Main Results:

  • Five new dimeric guaianolides (absinthins A-E) and seven known compounds were identified.
  • Absinthin C and isoanabsinthin demonstrated potent inhibition of nitric oxide production.
  • IC50 values for absinthin C and isoanabsinthin were determined as 1.52 μM and 1.98 μM, respectively.

Conclusions:

  • Artemisia absinthium is a rich source of structurally diverse dimeric guaianolides.
  • Absinthin C and isoanabsinthin possess significant anti-inflammatory properties.
  • These compounds represent potential therapeutic leads for inflammatory conditions.
