Background:
Conventional therapies for diseases that are associated with protein aggregation typically prevent rather than clear protein aggregates. We have proposed neutron dynamic therapy (NDT) as a physical clearance therapy for protein aggregates. Advanced glycation end-products (AGEs), which are aggregated proteins, have been implicated in diabetes, Alzheimer's, and heart disease. Herein, we report the use of the boron tracedrug UTX-51, under thermal neutron irradiation, as an NDT for the targeted clearance of glycated bovine serum albumin (Gly-BSA), a model of AGEs.
Materials And Methods:
Sodium dodecyl sulfate-polyacrylamide gel electrophoresis was performed to detect Gly-BSA decomposition by thermal neutron irradiation treated with UTX-51.
Results:
The combination of UTX-51 with neutron irradiation showed a decrease in band intensity of Gly-BSA.
Conclusion:
We present our NDT strategy, which has been used for the targeted clearance of Gly-BSA, suggesting that NDT with boron tracedrugs can be used for the treatment of AGEs-related disease.
Insights
Neutron dynamic therapy (NDT) with boron tracedrugs effectively clears glycated bovine serum albumin (Gly-BSA), a model for advanced glycation end-products (AGEs). This physical clearance therapy shows promise for treating AGEs-related diseases like diabetes and Alzheimer's.
Area of Science:
- Biochemistry
- Medical Physics
- Pharmacology
Background:
- Protein aggregation underlies diseases such as diabetes, Alzheimer's, and heart disease.
- Advanced glycation end-products (AGEs) are a key type of protein aggregate implicated in these conditions.
- Current therapies focus on preventing, not clearing, protein aggregates.
Purpose of the Study:
- To investigate neutron dynamic therapy (NDT) as a physical clearance method for protein aggregates.
- To evaluate the efficacy of a boron tracedrug, UTX-51, combined with thermal neutron irradiation for targeted clearance of AGEs.
- To use glycated bovine serum albumin (Gly-BSA) as a model for AGEs in this study.
Main Methods:
- Utilized sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to assess Gly-BSA decomposition.
- Applied thermal neutron irradiation in conjunction with the boron tracedrug UTX-51.
- Quantified the reduction in Gly-BSA levels based on band intensity in gel electrophoresis.
Main Results:
- The combination of UTX-51 and neutron irradiation resulted in a measurable decrease in Gly-BSA band intensity.
- This indicates successful decomposition and clearance of the Gly-BSA model protein aggregate.
- The results support the targeted clearance capability of NDT.
Conclusions:
- Neutron dynamic therapy (NDT) using boron tracedrugs offers a novel physical clearance strategy for protein aggregates.
- The study demonstrates successful targeted clearance of glycated bovine serum albumin (Gly-BSA) using this approach.
- NDT with boron tracedrugs holds potential for treating diseases associated with advanced glycation end-products (AGEs).
