Summary

This study presents a reproducible and affordable mouse model for critical osteochondral lesions (LOCs) in the trochlear groove. The model shows significant differences in repair between lesion and control areas, aiding future chondral pathology treatments.

Background:

  • Osteochondral lesions (OCLs) are challenging joint injuries.
  • Developing reliable animal models is crucial for studying OCLs and testing therapies.

Purpose of the Study:

  • To establish a reproducible and cost-effective critical osteochondral lesion (LOC) model in adult mice.
  • To evaluate the repair/regeneration capacity of this model.

Main Methods:

  • A 0.5mm diameter LOC was surgically created in the trochlear groove of 20 BKS mice.
  • Histological analysis using H&E and Safranin-O stains was performed at 7 and 14 days post-injury.
  • The International Cartilage Repair Society (ICRS) scale was used for evaluation, comparing lesion areas to healthy cartilage controls.

Main Results:

  • Significant histological differences were observed between the LOC area and control areas at both 7 and 14 days.
  • No significant differences in repair were detected between the 7-day and 14-day time points.

Conclusions:

  • The developed mouse model of LOC in the trochlear groove is highly reproducible.
  • This model can be utilized for future research aimed at developing improved treatments for chondral pathologies.
