Tumor-associated myeloid cells release transforming growth factor β (TGFβ), promoting cancer. We identified a novel network of delta-like 4 (Dll4), Notch, and TGFβ signals connecting cancer cells and myeloid cells for potential cancer therapy.
Area of Science:
- Oncology
- Cell Signaling
- Tumor Microenvironment
Background:
- Cell signals within the tumor microenvironment are crucial for cancer progression.
- Tumor-associated myeloid cells are known to secrete pro-tumorigenic factors, such as transforming growth factor β (TGFβ).
Purpose of the Study:
- To investigate the network of extrinsic signals linking malignant cells and tumor-infiltrating myeloid cells.
- To identify novel therapeutic targets within the tumor microenvironment.
Main Methods:
- Analysis of cell signaling pathways.
- Investigation of the interplay between malignant cells and myeloid cells.
Main Results:
- Discovery of a network involving delta-like 4 (Dll4), Notch, and TGFβ.
- This network connects malignant cells and tumor-infiltrating myeloid cells.
- This nexus represents a clinically relevant target for anticancer treatment.
Conclusions:
- A novel signaling network involving Dll4, Notch, and TGFβ has been identified.
- This network links cancer cells and myeloid cells within the tumor microenvironment.
- Targeting this nexus holds promise for developing new anticancer therapies.
