Insights

Telmisartan, an angiotensin receptor blocker (ARB), induces apoptosis in human renal cell carcinoma (RCC) cells by down-regulating Bcl-2 and activating caspase-3. This ARB did not induce apoptosis in non-cancerous renal cells, suggesting targeted anti-cancer effects.

Area of Science:

  • Oncology
  • Pharmacology
  • Molecular Biology

Background:

  • The renin-angiotensin system (RAS) regulates blood pressure but also promotes cancer cell proliferation.
  • Angiotensin receptor blockers (ARBs) can inhibit pro-survival signals in cancer by reducing anti-apoptotic molecules and caspase activity.

Purpose of the Study:

  • To evaluate the apoptotic effects of telmisartan, an ARB, on human renal cell carcinoma (RCC) and non-cancerous human renal (HEK) cell lines.
  • To investigate the mechanisms of telmisartan-induced apoptosis, including Bcl-2 and caspase-3 involvement.

Main Methods:

  • Human RCC (786) and HEK cells were treated with telmisartan (50-200 µM) or cisplatin (50 µM) for 4, 24, and 48 hours.
  • Apoptosis levels were assessed using MTT assays and flow cytometry.
  • Caspase-3 and Bcl-2 expression were analyzed via immunostaining.

Main Results:

  • Telmisartan significantly inhibited proliferation and induced apoptosis in RCC cells in a concentration- and time-dependent manner (P < 0.05).
  • Apoptosis induction in RCC cells was associated with positive caspase-3 staining and down-regulation of Bcl-2.
  • Telmisartan did not induce apoptosis in HEK cells (P > 0.05), while cisplatin promoted apoptosis in both cell types.

Conclusions:

  • Telmisartan demonstrates targeted apoptotic effects on human renal cell carcinoma cells.
  • The mechanism involves Bcl-2 down-regulation and caspase-3 activation, suggesting telmisartan's potential as an anti-cancer therapeutic agent.

The Intrinsic Apoptotic Pathway01:31

The Intrinsic Apoptotic Pathway

Internal cellular stress, such as cellular injury or hypoxia, triggers intrinsic apoptosis. The B-cell lymphoma 2 (Bcl-2) family of proteins are the primary regulators of the intrinsic apoptotic pathway. For example, during DNA damage, checkpoint proteins, such as Ataxia Telangiectasia Mutated (ATM protein) and Checkpoints Factor-2 (Chk2) proteins, are activated. These proteins phosphorylate p53 which further activates pro-apoptotic proteins, such as Bax, Bak, PUMA, and Noxa, and inhibits...

Antihypertensive Drugs: Angiotensin II Receptor Blockers01:30

Antihypertensive Drugs: Angiotensin II Receptor Blockers

In the renin-angiotensin-aldosterone system, a hormone called angiotensin II plays a crucial role. It binds to the AT1 receptors in vascular smooth muscles coupled with Gq proteins. The activation of these receptors activates an enzyme called phospholipase C, which releases two molecules: inositol trisphosphate and diacylglycerol. These molecules cause a chain reaction that leads to the phosphorylation of myosin light chains and promotes interaction between actin and myosin, leading to smooth...

Heart Failure Drugs: Inhibitors of Renin-Angiotensin System

The activation of the sympathetic nervous system and the renin-angiotensin-aldosterone system (RAAS) contributes to cardiac remodeling, and inhibiting the RAAS is a pharmacological target in heart failure management. As a result, neurohumoral modulation is a crucial treatment principle for managing heart failure. This approach involves using medications like ACE inhibitors (ACEIs), angiotensin receptor blockers (ARBs), β-blockers, mineralocorticoid receptor antagonists (MRAs), and neutral...

Cellular Injury V: Apoptosis and Autophagy

Cells respond to damage and stress through highly coordinated processes that decide whether they survive or undergo controlled self-destruction. Two major pathways involved in this regulation are apoptosis, a type of programmed cell death, and autophagy, a survival mechanism that helps cells adapt to adverse conditions.ApoptosisApoptosis removes aged or injured cells to maintain tissue balance. During this process, the cell shrinks, chromatin condenses and fragments, and membrane-bound...

Antihypertensive Drugs: Direct Renin Inhibitors

The renin-angiotensin-aldosterone system (RAAS) is an intricate physiological pathway involving numerous enzymes and hormones, including renin, angiotensin-converting enzyme (ACE), angiotensin I and II, and aldosterone. Imbalances within this system increase the production of angiotensin II and aldosterone. Increased angiotensin II levels promote vasoconstriction and blood pressure elevation. Concurrently, higher aldosterone levels stimulate sodium and water reabsorption in the kidneys,...

Targeted Cancer Therapies02:57

Targeted Cancer Therapies

The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against...
