  • Previous research in adults suggests amygdala dysfunction and altered frontal connectivity in depression, but adolescent-specific mechanisms remain understudied.

Purpose of the Study:

  • To investigate amygdala resting-state functional connectivity (RSFC) in adolescents diagnosed with MDD compared to healthy controls.
  • To explore the relationship between amygdala RSFC patterns and various symptom dimensions of depression in adolescents.
  • To utilize resting-state functional magnetic resonance imaging (fMRI) to identify neural network differences associated with adolescent depression.

Main Methods:

  • A cross-sectional study involving 41 adolescents and young adults (aged 12-19) with MDD and 29 age- and sex-matched healthy controls.
  • Whole-brain functional connectivity analysis using resting-state fMRI to assess correlations in spontaneous brain signal fluctuations.

  • Impaired amygdala-hippocampal/brainstem and amygdala-precuneus RSFC may be distinctive neurobiological features of adolescent MDD.
  • These altered circuits are implicated in memory processing, self-referential thought, and emotional regulation.
  • Findings suggest potential mechanisms for mood and vegetative symptoms in adolescent depression, possibly through disrupted processing of spontaneous memories and visceral signals.
