Background:

  • Pollen development in angiosperms is a complex process involving significant transcriptomic changes.
  • A switch from sporophytic to gametophytic gene expression programs occurs during male gametophyte development.

Purpose of the Study:

  • To identify and characterize novel genes with pollen-specific expression.
  • To validate the pollen-specific activity of identified gene promoters.
  • To demonstrate the utility of these promoters for targeted gene expression in pollen.

Main Methods:

  • Analysis of microarray databases to identify pollen-specific genes.
  • Validation of gene expression using Reverse Transcription Polymerase Chain Reaction (RT-PCR).
  • Construction of transcriptional fusions with the uidA reporter gene and the cytotoxin Barnase in Arabidopsis.

  • Five pollen-specific genes (POLLEN SPECIFIC GENE1-5) were selected.
  • Promoter activity was confirmed to be pollen-specific for at least two genes using reporter assays.
