Modified mesenchymal stromal cells (MSC) delivering tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) effectively target sarcoma cells, inducing apoptosis and inhibiting tumor growth. This novel approach shows promise for treating pediatric and young adult sarcomas.
Area of Science:
- Oncology
- Cancer Biology
- Stem Cell Therapy
Background:
- Sarcomas are aggressive pediatric and young adult cancers with high relapse and mortality rates.
- Mesenchymal stromal/stem cells (MSC) are implicated in sarcoma development and microenvironment support.
- Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a potent inducer of apoptosis.
Purpose of the Study:
- To investigate the therapeutic potential of genetically modified MSC to deliver TRAIL for sarcoma treatment.
- To evaluate the efficacy of MSC-TRAIL in inducing apoptosis and inhibiting tumor growth in various sarcoma types.
- To assess the antiangiogenic effects of MSC-TRAIL in an in vivo sarcoma model.
Main Methods:
- Gene modification of MSC to express TRAIL.
- In vitro co-culture experiments with osteosarcoma, rhabdomyosarcoma, and Ewing's Sarcoma (ES) cell lines.
- In vivo xenotransplantation model using ES to assess tumor apoptosis, size, and angiogenesis.
Main Results:
- MSC-TRAIL significantly induced apoptosis in all tested sarcoma cell lines.
- Sarcoma cell death was mediated by caspase-8 activation.
- In vivo, MSC-TRAIL demonstrated significant tumor apoptosis and antiangiogenic effects in ES xenografts.
- MSC-TRAIL persisted within the tumor stroma, indicating effective delivery.
Conclusions:
- Genetically modified MSC expressing TRAIL represent a promising therapeutic strategy for sarcomas.
- MSC-TRAIL exhibits dual mechanisms of action: direct tumor cell killing and antiangiogenesis.
- This approach offers novel therapeutic opportunities for challenging sarcoma histotypes.

Tumor Progression
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...
