Background:

  • Hereditary Cushing's syndrome is rare, often linked to adrenal dysplasia.
  • Cushing's disease (pituitary-dependent bilateral adrenal hyperplasia) is occasionally seen with Multiple Endocrine Neoplasia Type I.
  • No previous reports exist of Cushing's disease in multiple first-degree relatives.

Observation:

  • Two sisters presented with symptoms of hypercortisolism within an eight-month period.
  • Diagnostic studies confirmed Cushing's disease in both individuals.
  • Neither patient showed signs of other endocrine overactivity syndromes.

Findings:

  • Both sisters were diagnosed with ACTH-secreting pituitary adenomas.
  • Transsphenoidal surgery confirmed the pituitary adenomas.
  • This represents the first reported instance of Cushing's disease in first-degree relatives.

Implications:

  • Suggests a potential genetic predisposition for Cushing's disease.
  • Warrants further investigation into familial clustering of pituitary adenomas.
  • May influence diagnostic approaches for patients with a family history of endocrine disorders.
