Regulation of Expression at Multiple Steps01:23

Regulation of Expression at Multiple Steps

1.6K
The gene expression in cells is regulated at different stages: (i) transcription, (ii) RNA processing, (iii) RNA localization, and (iv) translation. Transcriptional regulation is mediated by regulatory proteins such as transcription factors, activators, or repressors—these control gene expression by initiating or inhibiting the transcription of genes. Once a precursor or pre-mRNA is produced, it undergoes post-transcriptional modification, including 5' capping, splicing, and the...

Regulation of Expression Occurs at Multiple Steps

27.5K
Gene expression can be regulated at almost every step from gene to protein. Transcription is the step that is most commonly regulated. This involves the binding of proteins to short regulatory sequences on the DNA. This association can either promote or inhibit the transcription of a gene associated with the respective sequence.
Transcription results in the generation of precursor (pre-mRNA) that consists of both exons and introns, which needs further processing before being translated to a...

Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

4.0K
Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl...

Role of Ephrin-Eph Signalling in Intestinal Stem Cell Renewal01:22

Role of Ephrin-Eph Signalling in Intestinal Stem Cell Renewal

2.8K
Erythropoietin-producing hepatocellular carcinoma receptor (Eph) and its ligand, Eph receptor-interacting protein (Ephrin) were first discovered in the human carcinoma cell line, hence the name. Ephrin-Eph interaction guides cells to reach their appropriate location in adult tissues. They also play an essential role in the immune system by helping in immune cell migration, adhesion, and activation. Based on their structure and function, Eph is divided into two classes — EphA and EphB.

Hedgehog Signaling Pathway02:33

Hedgehog Signaling Pathway

10.4K
The Hedgehog gene (Hh) was first discovered due to its control of the growth of disorganized, hair-like bristles phenotype in Drosophila, much like hedgehog spines. Hh plays a crucial role in the development of organs and the maintenance of homeostasis in both invertebrates and vertebrates. However, while Drosophila has only one Hh protein, mammals have multiple functional Hedgehog proteins - Sonic (Shh), Desert (Dhh), and Indian Hedgehog (Ihh). All of these homologous proteins have adapted to...

Summary

Chick early B-cell factor 1 (cEbf1) expression in somites is regulated by Shh signals from the notochord. This finding identifies cEbf1 as a medial sclerotome marker involved in somitogenesis.

Area of Science:

  • Developmental Biology
  • Molecular Biology
  • Genetics

Background:

  • Chick early B-cell factor 1 (cEbf1) is a transcription factor in the EBF family.
  • Previous studies showed Shh regulates cEbf1 expression in feathers.

Purpose of the Study:

  • To investigate if Shh signals from the notochord regulate cEbf1 expression in chick somites.
  • To detail the expression profile of cEbf1 during somitogenesis.

Main Methods:

  • Detailed analysis of cEbf1 expression in chick embryo somites (HH8-HH28).
  • Performed notochord ablation experiments.
  • Conducted Shh gain and loss of function experiments.

Main Results:

  • cEbf1 expression was primarily observed in the medial sclerotome and later around vertebral cartilage.
  • Experiments confirmed that Shh signaling from the notochord regulates medial somitic cEbf1 expression.

Conclusions:

  • cEbf1 is a medial sclerotome marker.
  • cEbf1 expression is downstream of and regulated by notochord-derived Shh.
  • cEbf1 may play a functional role in somitogenesis.
