Summary

Interferon-gamma (IFN-gamma) treatment in cancer patients caused a significant increase in cortisol levels, potentially impacting treatment effectiveness. This neuroendocrine response may be linked to fever during therapy.

Area of Science:

  • Endocrinology
  • Immunology
  • Oncology

Background:

  • Interferon-gamma (IFN-gamma) is used in cancer therapy.
  • The neuroendocrine effects of IFN-gamma are not fully understood.

Purpose of the Study:

  • To investigate the impact of IFN-gamma on cortisol and ACTH levels in patients with advanced metastatic tumors.
  • To explore the relationship between IFN-gamma administration, cortisol response, and fever.

Main Methods:

  • Plasma cortisol and ACTH levels were measured in 9 patients before and during IFN-gamma treatment.
  • Measurements were taken at specific time points after IFN-gamma administration.

Main Results:

  • A sharp rise in plasma cortisol levels was observed 2-4 hours after IFN-gamma administration, coinciding with fever onset.

Related Experiment Videos

  • Peak cortisol levels occurred 4-6 hours post-injection, while IFN-gamma serum levels peaked later (6-10 hours).
  • Conclusions:

    • Elevated cortisol during IFN-gamma treatment may represent a homeostatic response to the immunological stimulus.
    • High cortisol levels induced by IFN-gamma might potentially reduce its in vivo effectiveness in cancer patients.
