    Background:

    • Cleistocalycis Operculati Cortex is a key component in Compound Hibiscuse, used for antipruritic drugs.
    • Current quality control methods, like TLC, are insufficient for effective monitoring of Cleistocalycis Operculati Cortex.
    • There is a need for a more robust analytical method to ensure the quality of this medicinal bark.

    Purpose of the Study:

    • To establish and validate a reliable High-Performance Liquid Chromatography (HPLC) method for quantifying 3,3 -O-dimethylellagic acid in Cleistocalycis Operculati Cortex.
    • To provide a precise analytical tool for quality control of Cleistocalycis Operculati Cortex.
    • To address the limitations of existing quality assessment techniques.

    Main Methods:

    • Development of a reversed-phase HPLC method for the separation and determination of 3,3 -O-dimethylellagic acid.
    • Utilizing an external standard method for content calculation.
    • Validation of the method through linearity, recovery, and precision studies.

    Main Results:

    • The established HPLC method achieved effective separation of the target compound.
    • A good linear relationship was observed for 3,3 -O-dimethylellagic acid concentrations (1.00-25.0 mg/L), with a standard curve equation Y = 77.33X + 7.904 (r=0.9995).

    Conclusions:

    • This method offers strong technical support for enhancing the quality monitoring and control of Cleistocalycis Operculati Cortex.
    • Implementation of this method can ensure the consistent quality of raw materials for traditional Chinese medicine preparations.
