Low hydrostatic pressure (5 MPa) increases hypoxia-inducible factor-2α (HIF-2α) and cartilage-degrading enzymes in chondrocytes, independent of NF-κB. This suggests a role for hydrostatic pressure in osteoarthritis progression.
Area of Science:
- Biomedical Engineering
- Cell Biology
- Orthopedics
Background:
- Osteoarthritis (OA) progression is linked to Hypoxia-Inducible Factor-2α (HIF-2α).
- Previous studies suggest pressure amplitude affects HIF-2α in endothelial cells.
- The role of hydrostatic pressure in chondrocyte HIF-2α expression remains unclear.
Purpose of the Study:
- To investigate the effect of hydrostatic pressure on HIF-2α expression in articular chondrocytes.
- To determine if hydrostatic pressure influences genes associated with cartilage degradation.
- To explore the relationship between hydrostatic pressure, HIF-2α, and NF-κB signaling.
Main Methods:
- Articular chondrocytes were cultured and exposed to varying hydrostatic pressures (0, 5, 10, 50 MPa) and inflammatory stimuli.
- Gene expression of HIF-2α, HSP70, NF-κB, MMP-13, MMP-3, and VEGF was analyzed post-stimulation.
- Statistical analysis was performed to compare gene expression levels between groups.
Main Results:
- Inflammatory stress upregulated all tested genes.
- A hydrostatic pressure of 5 MPa significantly increased HIF-2α, MMP-13, and MMP-3 expression.
- At 5 MPa, HIF-2α increased independently of NF-κB; at 50 MPa, HIF-2α did not increase, while NF-κB and HSP70 did.
Conclusions:
- Hydrostatic pressure, particularly at 5 MPa, can regulate HIF-2α expression in chondrocytes.
- This regulation appears independent of NF-κB signaling at this pressure.
- Hydrostatic pressure may contribute to cartilage degeneration by inducing MMP-13 and MMP-3 via HIF-2α.
