    Summary

We developed a new image analysis method, two-dimensional sample entropy (SampEn2D), to capture pixel spatial distribution. SampEn2D reveals that elderly rat nerve images are more regular than young rat images.

    Area of Science:

    • Biomedical image analysis
    • Computational pathology
    • Neuroscience

    Background:

    • Traditional Shannon entropy for image analysis focuses solely on pixel gray levels.
    • The spatial distribution of pixels is crucial for comprehensive image analysis.
    • Sample entropy (SampEn) effectively quantifies time series irregularity but is not directly applicable to images.

    Purpose of the Study:

    • To introduce a novel 2D extension of sample entropy (SampEn2D) for image analysis.
    • To develop an entropy-based method that incorporates pixel spatial distribution for feature extraction.
    • To evaluate SampEn2D's utility in analyzing histological nerve images.

    Main Methods:

    • Extension of sample entropy (SampEn) to a two-dimensional algorithm (SampEn2D).

  • Application of SampEn2D to histological images of sural nerve segments from young and elderly rats.
  • Calculation of morphometric indexes (fiber count, area, perimeter) for comparison.
  • Main Results:

    • SampEn2D successfully extracts meaningful information from histological nerve images.
    • Elderly rat nerve images were classified as more regular (less complex) by SampEn2D compared to young rat images.
    • SampEn2D results are complementary to traditional morphometric indexes.

    Conclusions:

    • SampEn2D is a novel and effective method for image analysis, considering pixel spatial distribution.
    • The proposed method provides insights into age-related changes in nerve tissue complexity.
    • SampEn2D offers a complementary approach to existing morphometric analyses in histology.
