Bone Marrow Sampling and Transplants01:22

Bone Marrow Sampling and Transplants

2.8K
Bone marrow transplant is a potential cure for several diseases, including cancer and specific genetic disorders. Notably, this procedure is applicable for patients suffering from aplastic anemia, certain types of leukemia, severe combined immunodeficiency disease (SCID), Hodgkin's disease, non-Hodgkin's lymphoma, multiple myeloma, thalassemia, sickle-cell disease, and certain cancers.
The transplant begins with high doses of chemotherapy and radiation treatment, which aim to destroy...

Blood Transfusion and Agglutination

16.2K
Blood transfusion is a therapeutic measure to restore the blood volume after extensive blood loss due to an accident or a medical procedure. Blood transfusion involves drawing a certain amount of blood from a suitable donor and infusing it into the recipient.
History
The history of blood transfusion dates back to the 17th century, when early attempts were made in animals. In 1818 James Blundell, a British doctor, performed the first successful human blood transfusion. Later in 1900, Karl...

Stem Cell Therapy for Tissue Regeneration01:21

Stem Cell Therapy for Tissue Regeneration

5.0K
Stem cell therapy is a method used in regenerative medicine to repair and restore function to damaged tissues and organs. Stem cells have the potential to proliferate and differentiate into various tissue types, making them ideal candidates for tissue regeneration. For example, hematopoietic stem cell transplants are commonly used in blood cancer treatment to replenish damaged bone marrow and restore healthy blood cells.
Types of Stem Cells used in Stem Cell Therapy
The two main cell...

Summary

Eltrombopag shows promise for treating severe aplastic anemia (SAA) refractory to standard immunosuppressive therapy (IST). This thrombopoietin mimetic may directly stimulate bone marrow stem cells, offering a new treatment avenue for patients with bone marrow failure.

Area of Science:

  • Hematology
  • Immunology
  • Pharmacology

Background:

  • Standard immunosuppressive therapy (IST) for aplastic anemia, involving anti-thymocyte globulin (ATG) and cyclosporine, benefits two-thirds of patients.
  • A significant portion of IST responders experience relapse or persistent cytopenias, posing management challenges.
  • Previous attempts to improve outcomes with modified IST or hematopoietic cytokines have been unsuccessful.

Purpose of the Study:

  • To evaluate the efficacy of eltrombopag in patients with severe aplastic anemia (SAA) resistant to IST.
  • To explore the mechanism of action of eltrombopag in promoting hematopoietic recovery.
  • To investigate the potential of eltrombopag in combination therapies for SAA.

Main Methods:

  • A recent trial demonstrated the efficacy of eltrombopag, a thrombopoietin mimetic, in SAA patients refractory to IST.
  • Evidence suggests eltrombopag stimulates marrow stem and progenitor cells.
  • Ongoing institutional trials are exploring eltrombopag in upfront SAA treatment, IST-refractory SAA, and moderate aplastic anemia.

Main Results:

  • Eltrombopag has shown efficacy in treating severe aplastic anemia (SAA) that is refractory to standard immunosuppressive therapy (IST).
  • The drug appears to work by directly stimulating bone marrow stem and progenitor cells.
  • Ongoing studies are investigating eltrombopag's role in various SAA treatment settings.

Conclusions:

  • Eltrombopag represents a promising therapeutic option for patients with severe aplastic anemia (SAA) resistant to immunosuppressive therapy (IST).
  • Its mechanism of action, involving direct stimulation of hematopoietic stem cells, offers a novel approach to bone marrow failure treatment.
  • Further research through ongoing clinical trials is crucial to establish eltrombopag's role in upfront and refractory SAA management.
