    Background:

    • Archival samples offer valuable genetic insights for severe diseases like cancer.
    • Childhood acute lymphoblastic leukemia (ALL) presents unique challenges for genetic research.

    Purpose of the Study:

    • To assess the utility of archival bone marrow smears and biopsies for DNA extraction and genetic marker analysis.
    • To evaluate the impact of sample age and whole genome amplification (WGA) on DNA quality and genotyping success.

    Main Methods:

    • DNA extraction and purification from archival bone marrow samples (smears and biopsies).
    • Whole genome amplification (WGA) and analysis of short tandem repeats (STRs).
    • Genotyping of 33,683 single nucleotide polymorphisms (SNPs) using targeted next-generation sequencing.

    Main Results:

    • DNA concentration measurements varied between spectrophotometry and fluorometry.
    • STR detection rates decreased with longer fragments, especially after WGA.
    • Samples stored for 0-3 years yielded better results than those stored for 4-10 years.
    • Acceptable SNP genotyping call rates were achieved for 7 out of 42 archival samples.

    Conclusions:

    • Archival bone marrow samples are suitable for DNA extraction and multiple marker analysis in childhood ALL research.
    • WGA efficacy is limited, particularly with longer DNA fragments.
    • Optimized methods are needed for successful comprehensive SNP analysis from archival samples.
