    Background:

    • Effector T cell migration exacerbates tissue damage in inflammatory diseases, including graft-versus-host disease (GVHD).
    • Signaling pathways regulating T cell adhesion and migration downstream of chemokine receptors are not fully understood.
    • CRK and CRK-like (CRKL) adaptor proteins are implicated in cellular signaling, but their role in T cell migration is unclear.

    Purpose of the Study:

    • To investigate the role of CRK and CRKL adaptor proteins in effector T cell migration and adhesion.
    • To elucidate the signaling pathways involving CRK/CRKL in T cell trafficking.
    • To evaluate the therapeutic potential of targeting CRK proteins in GVHD models.

    Main Methods:

    • Utilized conditional knockout mice to study T cells deficient in CRK and CRKL.
    • Assessed integrin-dependent adhesion, chemotaxis, and diapedesis in CRK/CRKL-deficient T cells.

    • CRK proteins are critical mediators of T cell trafficking in inflammatory diseases.
    • CRK proteins represent potential therapeutic targets for mitigating GVHD while maintaining anti-leukemia immunity.
