Background:

The jun oncogene plays a role in cellular transformation.
Understanding the mechanisms of jun-mediated oncogenesis is crucial for cancer research.

Purpose of the Study:

To investigate the oncogenic transforming potential of the jun oncogene.
To determine the role of specific domains, particularly the transactivator domain, in jun-mediated transformation.
To explore the contribution of cellular jun to transformation induced by viral jun.

Main Methods:

Utilized viral constructs with terminal deletions of v-jun and c-jun.
Created recombinants between viral and cellular jun genes.
Assessed transformation efficiency of chicken embryo fibroblasts.

Main Results:

Cellular jun demonstrated low-efficiency transformation of fibroblasts.
High-efficiency transformation required alterations in the major transactivator domain.
A jun deletion lacking the transactivator domain was non-transforming.
Viral jun expression did not constitutively upregulate cellular jun in transformed cells.

Conclusions:

Transcriptional regulation is a significant factor in jun oncogenesis.
The transactivator domain is critical for high-efficiency jun-mediated transformation.
Cellular jun may not contribute to transformation when viral jun is highly expressed.
