Summary

Synthetic peptides targeting hormone mRNA sequences did not generate receptor-specific antibodies in this study. The synthesized "antisense" ACTH(1-24) peptide showed no receptor binding, despite inducing antibodies.

Background:

  • Synthetic peptides complementary to mRNA sequences can be used as antigens.
  • The goal is to generate receptor-specific antibodies for peptide hormones.

Purpose of the Study:

  • To synthesize an "antisense" peptide corresponding to human ACTH(1-24) mRNA.
  • To evaluate its antigenicity and the specificity of induced antibodies against the ACTH receptor.

Main Methods:

  • Synthesized an eikositetrapeptide "antisense" ACTH(1-24) (HTCAh).
  • Coupled HTCAh to carrier proteins (BSA, thyroglobulin) and immunized rabbits.
  • Tested antisera in binding and bioassays, and performed Western blots for receptor protein.

Main Results:

  • High-titer, specific antisera were generated against the HTCAh conjugate.
  • Antisera did not bind to MSH/ACTH receptor protein or affect ACTH/MSH binding assays.
  • HTCAh peptide showed low-affinity binding to a distinct site on melanoma cells, inducing melanin formation at high concentrations.

Conclusions:

  • The "antisense" peptide approach using HTCAh did not yield receptor-specific antibodies.
  • The generated antibodies were not specific for the ACTH receptor.
  • HTCAh peptide has independent biological activity on melanoma cells.
