Signaling lymphocytic activation molecule (SLAM) receptor CD244 is present on dendritic cells (DCs). Its absence in mice boosts pro-inflammatory cytokine production and enhances natural killer (NK) cell activation.
Area of Science:
- Immunology
- Cell Biology
Background:
- Signaling lymphocytic activation molecule (SLAM) receptors are crucial for immune responses, influencing NK cell cytotoxicity and cytokine production.
- The SLAM receptor CD244 (2B4, SLAMf4) is expressed on various immune cells, with its functions primarily studied in NK and T cells.
Purpose of the Study:
- To investigate the expression and function of CD244 in murine dendritic cell (DC) subsets.
- To determine the role of CD244 in DC maturation, cytokine production, and interaction with NK cells.
Main Methods:
- Analysis of CD244 expression across different murine DC subsets.
- Comparison of CD86 and MHC class II expression on DCs from CD244(-/-) and wild-type mice.
- Assessment of cytokine production by DCs upon Toll-like receptor (TLR) stimulation.
- Evaluation of NK cell activation by DCs in vitro.
Main Results:
- All examined murine DC subsets express CD244, with varying expression levels.
- DCs from CD244(-/-) mice showed reduced CD86 and MHC class II expression.
- Absence of CD244 led to significantly higher pro-inflammatory cytokine production by splenic DCs upon TLR stimulation (LPS and CpG).
- DCs from CD244(-/-) mice induced increased NK cell activation in vitro.
Conclusions:
- CD244 is expressed on all murine DC subsets and modulates their function.
- CD244 deficiency in DCs results in enhanced pro-inflammatory responses and increased NK cell activation.
- CD244 is identified as an immuno-modulatory receptor on dendritic cells.
