This study presents a novel workflow for analyzing lipoproteins, apoproteins, and lipids in serum using organic polymeric anion exchangers. The method offers high reproducibility for disease biomarker discovery and therapeutic applications.
Area of Science:
- Separation Science
- Analytical Chemistry
- Biochemistry
Background:
- Lipoproteins are crucial biomarkers for various diseases.
- Existing methods for lipoprotein analysis can be complex and time-consuming.
- A need exists for a comprehensive and reproducible workflow for lipoprotein analysis.
Purpose of the Study:
- To develop and validate a novel workflow for the comprehensive analysis of lipoproteins, apoproteins, and lipids from a single serum sample.
- To utilize organic polymeric anion exchangers for efficient lipoprotein enrichment and separation.
- To integrate mass spectrometry techniques for detailed apoprotein and lipid profiling.
Main Methods:
- Synthesis and derivatization of organic polymers (poly(GMA/DVB), poly(GMA/EGDMA), poly(GPE/DVB)) as strong anion exchangers.
- Enrichment of lipoproteins via interaction with phosphate groups on their surface at pH 7.5.
- Isolation of high-density lipoproteins (HDL) by precipitating very low-density lipoproteins (VLDL) and low-density lipoproteins (LDL) using phosphotungstic acid.
- Delipidation using liquid/liquid extraction.
- Apoprotein profiling using Matrix-Assisted Laser Desorption/Ionization-Mass Spectrometry (MALDI-MS).
- Lipid analysis using gold nanoparticles in Laser Desorption/Ionization-Mass Spectrometry (LDI-MS).
- Implementation of a solid-phase extraction (SPE)-based methodology.
Main Results:
- Successful synthesis and application of chemically stable organic polymeric anion exchangers for lipoprotein enrichment.
- Demonstrated effective separation and enrichment of lipoproteins based on their surface charge.
- Achieved detailed apoprotein profiling using MALDI-MS and lipid analysis using LDI-MS from a single serum sample.
- Reported the first solid-phase extraction-based methodology for lipoproteomics.
Conclusions:
- The developed workflow enables comprehensive analysis of intact lipoproteins, apoproteins, and lipids with high reproducibility.
- This lipoproteomics workflow offers significant advantages in terms of efficiency and detail compared to existing methods.
- The workflow holds clinical significance for disease diagnosis and therapeutic monitoring due to the role of lipoproteins as biomarkers.
