Background:

  • Alzheimer's disease (AD) is a neurodegenerative disorder.
  • Humanin ([Gly14]-Humanin, HNG) is a peptide with potential neuroprotective properties.
  • The streptozotocin (STZ) model mimics sporadic AD by disrupting insulin signaling.

Purpose of the Study:

  • To investigate if humanin protects against STZ-induced spatial memory deficits.
  • To determine if humanin preserves hippocampal PI3/Akt signaling after STZ administration.

Main Methods:

  • Rats received bilateral ventricular injections of STZ to model sporadic AD.
  • Humanin or saline was administered for 10 days post-STZ.
  • Spatial memory was assessed using the Morris water maze.
  • Hippocampal phosphorylated Akt (pAkt) levels were measured via Western blot.

Main Results:

  • STZ significantly impaired spatial memory in rats.
  • Humanin, across various doses, did not restore STZ-induced memory deficits.
  • Humanin failed to prevent the disruption of hippocampal pAkt levels.

Conclusions:

  • Humanin is ineffective in ameliorating memory impairment in this STZ-induced sporadic AD model.
  • Humanin does not restore the disrupted insulin signaling pathway (PI3/Akt) in the hippocampus.
  • The neuroprotective effects of humanin may not extend to conditions involving insulin signaling disruption.
