Congenital kidney and urinary tract anomalies (CAKUT) cause pediatric renal failure. We developed an integrated disector method, a faster, cost-effective digital approach to accurately quantify nephron number, aiding research into kidney disease origins.
Area of Science:
- Nephrology
- Developmental Biology
- Medical Imaging
Background:
- Congenital Anomalies of the Kidney and Urinary Tract (CAKUT) are a primary cause of pediatric renal failure.
- Reduced nephron number is linked to adult hypertension and kidney disease.
- Accurate nephron quantification is crucial for understanding kidney development and disease.
Purpose of the Study:
- To present an updated, digitized method for accurate nephron number quantification.
- To provide a rapid, cost-effective alternative to traditional stereological methods.
- To facilitate research into the origins of human kidney disease using mouse models.
Main Methods:
- Developed the integrated disector method, a digitized version of the physical disector/fractionator technique.
- Utilized free, open-source Fiji software for stereological measurements.
- Applied the method to embryonic and post-natal mouse kidneys.
Main Results:
- The integrated disector method accurately quantitates nephron number.
- This digital approach is significantly faster and more cost-effective than the traditional method.
- The method is adaptable for stereological analysis in other organ systems.
Conclusions:
- The integrated disector method offers an accurate, efficient, and accessible tool for nephron counting.
- This advancement aids research into CAKUT and the long-term consequences of low nephron endowment.
- The method's adaptability broadens its potential applications in biological research.
