Tumor progression is a phenomenon where the pre-formed tumor acquires successive mutations to become clinically more aggressive and malignant. In the 1950s, Foulds first described the stepwise progression of cancer cells through successive stages.

Summary

ADAMTS-4 and ADAMTS-5 are overexpressed in colorectal cancer, correlating with disease progression. Conversely, anti-angiogenic ADAMTS-1 and ADAMTS-20 are downregulated, suggesting a role in cancer invasion.

Background:

  • ADAMTSs (a disintegrin and metalloproteinase with thrombospondin motifs) are secreted proteinases influencing cell functions and angiogenesis.
  • Their role in colorectal cancer (CRC) progression and invasion remains to be fully elucidated.

Purpose of the Study:

  • To investigate the expression and potential role of ADAMTS family members in colorectal cancer progression and invasion.

Main Methods:

  • Quantitative real-time PCR (RT-PCR) to assess mRNA expression of ADAMTS-1, -4, -5, and -20 in CRC tissues and cell lines.
  • Western blot analysis to confirm protein expression and detect degradation.
  • Immunohistochemical analysis to determine in situ expression and localization of ADAMTS proteins.

Main Results:

  • ADAMTS-4 and ADAMTS-5 showed significant overexpression in CRC tissues and cell lines, correlating with advanced cancer stage and aggressiveness.
