    Area of Science:

    • Cardiovascular Physiology
    • Pharmacology

    Background:

    • Adult rat ventricular myocytes exhibit Ca2+-tolerant contractions.
    • Myocyte contraction frequency (fm) is temperature-dependent.

    Purpose of the Study:

    • To investigate the effect of verapamil on myocyte contraction frequency across different temperatures.
    • To assess the implications for cardiac surgery under hypothermia.

    Main Methods:

    • Electrical stimulation of isolated rat ventricular myocytes.
    • Determination of maximal contraction frequency (fm) at varying temperatures.
    • Analysis of fm using Arrhenius plots in the presence and absence of verapamil.

    Main Results:

    • In drug-free solution, fm followed the Arrhenius equation from 7°C to 39.5°C.
    • Verapamil induced a discontinuity in the Arrhenius plot of fm around 27°C.
    • Verapamil's inhibitory effect on fm was more pronounced above 27°C than below.

    Conclusions:

    • Verapamil exhibits temperature-dependent effects on myocyte contractility.
    • Findings suggest potential implications for using calcium antagonists in cardiac surgery during deep hypothermia.
