SOX11 gene expression is epigenetically regulated by DNA methylation and histone modifications in cancer. Histone deacetylase inhibitors can re-express SOX11 in cancer cells with low promoter methylation, offering potential therapeutic strategies.
Area of Science:
- Epigenetics
- Cancer Biology
- Molecular Oncology
Background:
- SOX11, a neural transcription factor, plays a role in tumorigenesis and exhibits precise transcriptional regulation.
- Investigating epigenetic differences in SOX11 expression between normal and cancerous cells is crucial for understanding its role in malignancy.
Purpose of the Study:
- To explore epigenetic regulation of SOX11 expression in normal versus neoplastic cells.
- To investigate the potential of re-expressing SOX11 using histone deacetylase (HDAC) or EZH2 inhibitors.
Main Methods:
- Assessed DNA methylation using bisulfite sequencing and pyrosequencing.
- Evaluated H3K27me3 enrichment via ChIP-qPCR.
- Utilized HDAC inhibitors (Vorinostat, trichostatin A) to induce SOX11 expression.
Main Results:
- Non-malignant cells show low SOX11 promoter methylation and high H3K27me3.
- B-cell lineage cancers exhibit de novo SOX11 promoter methylation; solid tumors show variable methylation.
- HDAC inhibitors successfully induced SOX11 expression in cancer cells with low SOX11 promoter methylation.
Conclusions:
- SOX11 is epigenetically regulated by repressive histone marks in normal cells.
- Neoplastic SOX11 regulation involves complex DNA methylation and histone modifications.
- Re-expressing SOX11 in non-methylated cancer cells via HDAC inhibitors is clinically relevant and demonstrated efficacy.
