    Summary

    Persistent facial pain in a rodent model impairs self-regulation and adaptation to environmental demands. This study demonstrates that chronic constriction injury of the infraorbital nerve (CCI-ION) affects behavioral regulation in rats.

    Background:

    • Self-regulation is a complex cognitive function crucial for adapting to environmental demands.

    • Chronic pain conditions are known to affect cognitive functions, but their impact on self-regulation requires further elucidation.

    Purpose of the Study:

    • To establish a rodent model for studying self-regulation.
    • To determine if persistent facial pain, induced by chronic constriction injury of the infraorbital nerve (CCI-ION), influences self-regulatory behaviors.

    Main Methods:

    • Thirty male Sprague-Dawley rats were used, divided into CCI-ION and naïve control groups.
    • A two-part behavioral paradigm assessed self-regulation: a cued go/no-go task and a persistence trial.
    • Behavioral testing occurred over 5 weeks post-surgery, with data analyzed using t tests and ANOVA.

    Main Results:

    • CCI-ION surgery successfully induced persistent mechanical hypersensitivity in the whisker pad.
    • Naïve rats showed a significant decrease in lever presses during the persistence task post-manipulation.
    • CCI-ION rats did not exhibit this adaptive decrease in lever pressing, indicating impaired behavioral regulation.

    Conclusions:

    • Persistent pain significantly influences behavioral regulation in a rodent model.
    • Animals with persistent pain may exhibit difficulties in adapting to changing environmental demands.
    • The findings suggest a link between chronic pain and deficits in cognitive control and behavioral flexibility.
