Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens.
The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper. Background:

  • 10% formalin is the standard fixative for tissue preservation.

Purpose of the Study:

  • To evaluate common, readily available solutions as potential transit fixatives for tissue specimens.
  • To determine the efficacy of these alternative solutions in preserving tissue architecture for a limited period.
  • To identify viable substitutes for formalin as a temporary transport medium.

Main Methods:

  • Commonly available solutions including Spirit, Saline, Betadine, Hydrogen peroxide, Local anesthesia, Rose water, Coconut oil, Coconut water, Ice cold water, Honey, and Milk were tested.
  • Formalin served as the control fixative.
  • Fresh tissue samples were divided into small pieces and placed in each test solution for 8 hours before transfer to formalin.
