Area of Science:

  • Molecular Biology
  • Cell Biology
  • Genetics

Background:

  • Microdissection enables pure cell isolation for mutation analysis but often compromises cellular morphology visualization.
  • Existing methods lack sufficient resolution for detailed morphological assessment of microdissected cells.

Purpose of the Study:

  • To develop and evaluate the MultiTech DNA extraction solution for improved cell morphology visualization during microdissection.
  • To assess the solution's efficacy in preserving cell morphology, DNA quality, and mutation detection capabilities.

Main Methods:

  • 121 cytological samples (fine-needle aspirates, sputum, pleural fluid, bronchial washings) were analyzed.
  • DNA extracted from microdissected cells underwent epidermal growth factor receptor (EGFR) mutation analysis using pyrosequencing, Sanger sequencing, and PNA-mediated real-time PCR clamping.
  • Morphological features, DNA quality, and quantity were assessed to evaluate the MultiTech solution's performance.

Main Results:

  • The MultiTech solution improved morphology of stained cells prior to microdissection.
  • It yielded more DNA than the QIAamp DNA Mini Kit in samples with minimal cell counts (25-50 cells).
  • Identical EGFR mutations were detected across different platforms, even in samples with as few as 25 microdissected cells.

Conclusions:

  • The MultiTech DNA extraction solution reliably enhances cell morphology resolution for microdissection.
  • It is particularly effective for EGFR mutation analysis in samples with limited cellular material.
