High expression of XB130 adaptor protein is linked to poorer prognosis and increased metastasis in osteosarcoma (OS) patients. This finding suggests XB130 could be a valuable prognostic marker and therapeutic target for OS.
Area of Science:
- Oncology
- Molecular Biology
- Biochemistry
Background:
- Identifying prognostic factors for osteosarcoma (OS) is crucial for tailoring aggressive treatment strategies.
- XB130, an adaptor protein, is a known prognostic factor in other cancers, but its role in OS is unexplored.
Purpose of the Study:
- To investigate the association between XB130 expression and clinicopathologic features, prognosis, and metastatic potential in osteosarcoma.
- To explore the functional role of XB130 in OS cell invasion and metastasis in vitro and in vivo.
Main Methods:
- Retrospective immunohistochemical analysis of XB130 expression in 60 osteosarcoma and noncancerous bone tissue pairs.
- Correlation of XB130 expression with clinicopathological parameters and patient survival.
- Assessment of XB130 silencing effects on OS cell invasion (in vitro) and lung metastasis (in vivo) using RNA interference.
Main Results:
- XB130 expression was significantly higher in OS tissues compared to noncancerous tissues (P=0.001).
- High XB130 expression correlated with advanced clinical stage (P=0.002) and distant metastasis (P=0.001).
- Patients with high XB130 expression exhibited significantly shorter overall and disease-free survival (P<0.001).
- XB130 depletion inhibited OS cell invasion and lung metastasis.
- High XB130 expression and distant metastasis were identified as independent poor prognostic factors.
Conclusions:
- XB130 overexpression is associated with poor prognosis and increased metastatic potential in osteosarcoma.
- XB130 may serve as a prognostic biomarker for optimizing clinical treatment strategies in OS patients.
- XB130 represents a potential molecular target for novel osteosarcoma therapies.
