Background:

  • Hydrophobic interaction chromatography (HIC) is a key protein purification method.
  • HIC relies on salting-out salts to enhance hydrophobic interactions.
  • Ammonium sulfate, a common HIC salt, is unsuitable for alkaline pH conditions.

Purpose of the Study:

  • To evaluate monosodium glutamate (MSG) as a salt for HIC at alkaline pH (9.5).
  • To assess the binding and elution characteristics of proteins using MSG in HIC.
  • To determine MSG's efficacy in protein purification and separation.

Main Methods:

  • Proteins (ribonuclease A, ovalbumin, beta-lactoglobulin) were applied to a phenyl superose column.
  • A 2 M concentration of monosodium glutamate was used as the salt at pH 9.5.
  • Proteins were eluted by decreasing the salt concentration.
  • Other salts like guanidine, sodium sulfate, and sodium aspartate were also examined.
