Secretogranin-1 (Sg1) is elevated in early multiple sclerosis (MS) patients. This finding offers potential for improved early diagnosis and understanding of MS pathogenesis.

Background:

  • Multiple sclerosis (MS) is a chronic central nervous system (CNS) inflammatory disease with an unknown etiology.
  • Identifying proteins with altered cerebrospinal fluid (CSF) abundance in MS patients can offer insights into pathogenesis and aid in diagnosis and prognosis.

Purpose of the Study:

  • To discover proteins with differential CSF abundance between relapsing-remitting MS (RRMS) patients and neurological controls.
  • To identify potential biomarkers for MS diagnosis and prognosis.

Main Methods:

  • Utilized iTRAQ labeling and Orbitrap mass spectrometry to quantify proteins in CSF from RRMS patients and controls.
  • Employed multiple reaction monitoring (MRM) for verification of candidate biomarkers in an independent patient cohort.

Main Results:

  • Seven protein candidates were identified, including chitinase-3-like protein 1 and secretogranin-1 (Sg1).
  • Secretogranin-1 (Sg1) was found to be significantly increased in early-MS patients compared to both RRMS patients and neurological controls.

Conclusions:

  • Secretogranin-1 (Sg1) shows potential as a biomarker for early-stage MS.
  • Further research is warranted to elucidate Sg1's role in early MS pathogenesis and its utility as a diagnostic marker.
