Pharmacogenetics of Drug Targets: β₂-Adrenergic Receptors, Apo E, Thymidylate Synthase01:11

Pharmacogenetics of Drug Targets: β₂-Adrenergic Receptors, Apo E, Thymidylate Synthase

69
Genetic polymorphisms in drug targets have emerged as critical determinants of interindividual variability in drug response and toxicity. Pharmacogenomic investigations increasingly focus on identifying these variations to personalize and optimize therapeutic interventions. A drug target may be a receptor, enzyme, or signaling protein involved in pharmacologic responses or disease-related pathways. While early pharmacogenetic studies focused primarily on drug metabolism, current research...

Pharmacogenomics: Identification of New Drug Targets

86
Advances in genomics have profoundly influenced drug discovery by increasing both the speed and accuracy of pharmaceutical development. Pharmacogenomics, which examines how genetic variation influences drug response, facilitates the identification of novel therapeutic targets and enables patient stratification for personalized treatment. These strategies contribute to improved drug efficacy, minimized adverse effects, and more efficient clinical trial design.Mapping genetic differences...

Pharmacogenetic Phenotypes: Alterations in Pharmacokinetics, Drug Targets and Biologic Milieu

116
Genetic variations significantly influence drug response through pharmacokinetics, receptor interactions, and biologic milieu modifications. Pharmacokinetic alterations impact drug metabolism and clearance, affecting efficacy and toxicity. Variants in drug-metabolizing enzymes, such as CYP2C9 and CYP2C19, alter drug activation and elimination. For example, CYP2C9 loss-of-function variants require lower warfarin doses to prevent excessive bleeding, while CYP2C19 variants reduce clopidogrel...

Diabetes Mellitus: Type 2 and Gestational

5.6K
Type 2 diabetes, characterized by insulin resistance, arises when the insulin receptors on cells lose responsiveness to insulin, diminishing the cell's capacity to take up glucose, resulting in elevated blood glucose levels. To receive a diagnosis of Type 2 diabetes, a series of blood glucose tests are necessary to assess whether the blood glucose falls within normal parameters. If the result is out of the normal range, a patient may be diagnosed as prediabetic or diabetic, depending on the...

Principles of Pharmacogenetics: Types of Genetic Variants

102
The human genome is over 99.9% identical between individuals, yet genetic differences exist at millions of bases. The human genome contains approximately 3 million variant positions per individual, many of which are heterozygous, contributing to genetic diversity and individual traits. Genetic variations include single-nucleotide polymorphisms (SNPs), insertions, deletions, and copy number variations (CNVs).SNPs, the most common variation, involve single-base changes in DNA. These can be...

Pharmacogenetics of Drug Metabolism: Overview

131
Genetic polymorphism in drug metabolism is crucial to the inter-individual variability observed in drug responses. Drug metabolism primarily involves the chemical modification of drugs and other xenobiotics to enhance their elimination by increasing their polarity. Two main classes of enzymes mediate this biotransformation process: Phase I enzymes, primarily cytochrome P450s, catalyze oxidation and reduction reactions, while other enzymes, such as esterases, mediate hydrolysis, and Phase II...

Summary

Adiponectin gene (ADIPOQ) polymorphisms are linked to type 2 diabetes mellitus (T2DM) in Chinese individuals. Specific single nucleotide polymorphisms (SNPs) and haplotypes show significant associations with T2DM risk.

Area of Science:

  • Genetics and Molecular Biology
  • Endocrinology and Metabolism
  • Population Health

Background:

  • Type 2 diabetes mellitus (T2DM) is a significant global health concern.
  • Adiponectin, a hormone involved in glucose regulation and fatty acid breakdown, is encoded by the ADIPOQ gene.
  • Genetic variations in ADIPOQ may influence T2DM susceptibility.

Purpose of the Study:

  • To investigate the association between ADIPOQ gene polymorphisms and T2DM in a Chinese population.
  • To identify specific SNPs and haplotypes within the ADIPOQ gene that are risk factors for T2DM.

Main Methods:

  • A case-control study was conducted with 340 T2DM patients and 340 normal glucose tolerant (NGT) controls.
  • Three single nucleotide polymorphisms (SNPs): rs182052, rs1501299, and rs7627128 in the ADIPOQ gene were genotyped using TaqMan methods.
  • Haplotype analysis was performed to assess combinations of SNPs.

Main Results:

  • All three genotyped SNPs (rs7627128, rs1501299, and rs182052) demonstrated a significant association with T2DM.
  • The A-A-T haplotype was found to be more frequent in T2DM patients (OR = 2.10; 95% CI: 1.44-2.90; p < 0.001).
  • Conversely, the G-A-T haplotype was more prevalent in the control group (OR = 0.66; 95% CI: 0.54-0.81; p < 0.001).

Conclusions:

  • Genetic polymorphisms in the ADIPOQ gene are significantly associated with type 2 diabetes.
  • Specific ADIPOQ haplotypes may serve as genetic markers for T2DM risk in the Chinese population.
