p53 codon 72 polymorphism and hepatocellular carcinoma: a meta-analysis.

This meta-analysis indicates that the p53 codon 72 Pro/Pro genotype is linked to a higher risk of hepatocellular carcinoma (HCC) in Caucasian populations. Further global research is recommended to confirm this association between p53 polymorphism and HCC.

Background:

Purpose of the Study:

- A systematic literature search of PubMed was performed to identify relevant studies.
- Studies with available genotype frequencies for the p53 Arg72Pro polymorphism were included.
- Pooled odds ratios (OR) with 95% confidence intervals (CI) were calculated to assess the association.

- Ten studies comprising 1,371 HCC cases and 2,517 controls were analyzed.
- Overall, variant genotypes (Pro/Pro) were associated with an increased HCC risk (OR 1.355, p=0.024).
- Stratified analysis revealed a significantly increased HCC risk in Caucasians with the Pro/Pro genotype under a recessive model (OR 1.927, p=0.017).
- No association was found between the p53 codon 72 polymorphism and HBV/HCV-positive HCC.
- Publication bias was assessed and found to be minimal.
Conclusions:
- The p53 codon 72 Pro/Pro genotype is associated with an elevated risk of HCC in Caucasian individuals.
- Further large-scale international studies are necessary to validate this association and explore the polymorphism's role in HCC development.
