Summary

Computer-assisted sperm analysis (CASA) accuracy varies by counting chamber. Disposable chambers are suitable for routine use, but plain slides and tissue culture dishes also perform well with proper quality control.

Semen samples were analyzed using a computer-assisted semen analyzer (CASA).

Sperm concentration and motility were measured and compared across chamber types.

Main Results:

The reusable Makler chamber yielded significantly higher sperm concentration counts compared to other chambers.
Sperm motility was significantly higher in drop-loaded chambers versus capillary-loaded chambers.
A plain glass slide and tissue culture dish cover demonstrated comparable or better performance in semen assessment.

Conclusions:

Disposable counting chambers are suitable for routine semen analysis with CASA.
Plain glass slides and tissue culture dish covers are acceptable alternatives with proper quality control.
Specifying the counting chamber type in test reports is recommended for clarity and consistency.
