Area of Science:

  • Andrology
  • Reproductive Medicine
  • Laboratory Diagnostics

Background:

  • Computer-assisted semen analysis (CASA) is crucial for andrology laboratories.
  • Accurate sperm counting and motility assessment are vital for male fertility evaluation.
  • Different counting chambers exist for semen analysis, impacting results.

Purpose of the Study:

  • To evaluate the accuracy and precision of various counting chambers used with CASA systems.
  • To compare sperm concentration and motility results obtained from different chamber types.
  • To determine the suitability of disposable vs. reusable chambers for routine CASA.

Main Methods:

  • A quality-control study comparing sperm analysis using different counting chambers: Makler chamber, GoldCyto chamber, Leja chamber, plain glass slide, and tissue culture dish cover.

  • Semen samples were analyzed using a computer-assisted semen analyzer (CASA).
  • Sperm concentration and motility were measured and compared across chamber types.
  • Main Results:

    • The reusable Makler chamber yielded significantly higher sperm concentration counts compared to other chambers.
    • Sperm motility was significantly higher in drop-loaded chambers versus capillary-loaded chambers.
    • A plain glass slide and tissue culture dish cover demonstrated comparable or better performance in semen assessment.

    Conclusions:

    • Disposable counting chambers are suitable for routine semen analysis with CASA.
    • Plain glass slides and tissue culture dish covers are acceptable alternatives with proper quality control.
    • Specifying the counting chamber type in test reports is recommended for clarity and consistency.
