Summary
Three shrimp Rab proteins (LvRab5B, LvRab6A, LvRab7) were identified and their expression analyzed. Shrimp immune response to viral infection involves these Rab genes, showing significant up-regulation after virus challenge.
Area of Science:
- Molecular Biology
- Immunology
- Marine Biology
Background:
- Rab proteins are GTP-binding proteins crucial for membrane trafficking.
- Understanding shrimp immune responses is vital for aquaculture.
Purpose of the Study:
- To clone and characterize three Rab genes (LvRab5B, LvRab6A, LvRab7) from Litopenaeus vannamei.
- To investigate the role of these Rab genes in shrimp immune response to viral infections.
Main Methods:
- Cloning of full-length cDNA sequences for LvRab5B, LvRab6A, and LvRab7.
- Quantitative reverse transcription polymerase chain reaction (qRT-PCR) to analyze mRNA expression levels.
- Challenging shrimp with infectious hypodermal and hematopoietic necrosis virus and white spot syndrome virus.
Main Results:
- Full-length cDNA sequences of LvRab5B (1383 bp), LvRab6A (873 bp), and LvRab7 (767 bp) were obtained.
- mRNA expression levels of LvRab5B, LvRab6A, and LvRab7 were significantly increased in shrimp hepatopancreas post-viral challenge.
- These findings suggest a role for these Rab proteins in the shrimp antiviral immune defense.
Conclusions:
- The cloned Rab genes, LvRab5B, LvRab6A, and LvRab7, are expressed in Litopenaeus vannamei.
- These genes are involved in the shrimp's immune response to viral pathogens.
- Further research can explore the specific mechanisms of these Rab proteins in antiviral immunity.

Rab Proteins
5.4K
Rab proteins constitute the largest family of monomeric GTPases, of which 70 members are present in humans. Rab proteins and their effectors regulate consecutive stages of vesicle transport such as vesicle transport, docking, and fusion to the correct recipient membrane.
Rab proteins switch between a cytosolic, GDP-bound inactive state and a membrane-anchored, GTP-bound active state. By themselves, Rabs show slow rates of GDP/GTP exchange and GTP hydrolysis. Thus, Rab proteins are considered...

Rab Cascades
3.8K
Rab GTPases act in a regulated cascade during membrane fusion, helping the lipid bilayers mix. The Rab family of proteins are active when bound to GTP, and inactive when bound to GDP. Hence, they act as guanine nucleotide-dependent molecular switches. Rab-GTP recognizes and binds to long or short-range tethering proteins to capture the target vesicle. These tethers coordinate with SNAREs on the vesicle and the target membrane to assemble the trans SNARE complex that locks the mixing bilayers.
