Summary
This study isolated and identified seven proanthocyanidins (PAs) from Salix daphnoides bark. Differences in flavan-3-ol composition and chain length were observed in polymeric PA fractions.
Area of Science:
- Phytochemistry
- Natural Product Chemistry
Background:
- Salix daphnoides bark is a potential source of bioactive compounds.
- Proanthocyanidins (PAs) are a class of polyphenols with diverse biological activities.
Purpose of the Study:
- To isolate and characterize proanthocyanidins from Salix daphnoides bark.
- To investigate the structural differences between regular and unusual polymeric PA fractions.
Main Methods:
- Aqueous-methanolic extraction of Salix daphnoides bark.
- Chromatographic separation techniques including Sephadex® LH-20, MCI®, Diol, and RP-18.
- Spectroscopic analysis: Mass spectrometry, 1D- and 2D-NMR, 13C-NMR, Circular Dichroism, Polarimetry.
- Chemical degradation methods: Thiolysis, acid hydrolysis, phloroglucinol degradation.
- Centrifugal Partition Chromatography (CPC) for enrichment of specific fractions.
Main Results:
- Seven proanthocyanidins were isolated and identified: Procyanidin B1, B2, B3, B4, C1, and two extended oligomers (epicatechin-(4β→8)-epicatechin-(4β→8)-catechin and epicatechin-(4β→8)-epicatechin-(4β→8)-epicatechin-(4β→8)-catechin).
- Two distinct fractions of polymeric flavan-3-ols were characterized: 'regular' polymeric PAs and 'unusual' PAs.
- Differences in flavan-3-ol unit composition and middle chain length were observed between the two polymeric PA fractions.
Conclusions:
- Salix daphnoides bark is a rich source of diverse proanthocyanidins.
- The study highlights structural variations within polymeric proanthocyanidins, suggesting differential biosynthesis or degradation pathways.
- Further investigation into the biological activities of these isolated compounds is warranted.
