Area of Science:

  • Biochemistry
  • Polymer Science
  • Microbiology

Background:

  • Levan, a fructan polymer, has diverse industrial applications.
  • Levan's utility is often dictated by its molecular weight.
  • Molecular weight is typically influenced by enzyme source and reaction conditions.

Purpose of the Study:

  • Investigate the effect of enzyme concentration on levan molecular weight distribution.
  • Specifically examine levansucrase from Bacillus subtilis 168.
  • Determine if enzyme concentration is an independent factor controlling levan characteristics.

Main Methods:

  • Utilized levansucrase from Bacillus subtilis 168.
  • Varied enzyme concentrations (0.01 μM, 0.1 μM, and >0.5 μM).
  • Analyzed the resulting levan molecular weight distributions.

Main Results:

  • A bimodal levan distribution was observed at standard enzyme concentrations (0.1 μM).
  • High enzyme concentrations (>0.5 μM) exclusively produced low molecular weight levan with a normal distribution.
  • Low enzyme concentrations (0.01 μM) synthesized high molecular weight levan with a normal distribution.

Conclusions:

  • Enzyme concentration is a key determinant of levan molecular weight distribution for Bacillus subtilis levansucrase.
  • Controlling enzyme concentration allows for targeted synthesis of specific levan molecular weights.
  • This finding offers a method to tailor levan properties for various industrial applications.
